NEK7: An Essential Regulator of NLRP3 Inflammasome Activation

NEK7 is the smallest of the eleven (NEK1-NEK11) mammalian NIMA-related kinase (NEK) family serine/threonine kinases and is expressed in many tissues. Its importance in mitotic regulation and NLRP3 inflammasome activation has been established, with inflammasome activation involving NEK7’s direct binding to NLRP3. Inflammasome activation is associated with pyroptosis and mediates the release of pro-inflammatory cytokines IL-1β and IL-18 and the cleavage of pore-forming gasdermin D. Pyroptosis is linked to many inflammatory diseases, and thus therapeutic targeting of NEK7-mediated inflammasome activation continues to be an exciting field of research (1).
