Background

This gene product is a component of a nucleolar small nuclear ribonucleoprotein (snRNP) particle thought to participate in the first step in processing preribosomal RNA. It is associated with the U3, U8, and U13 small nuclear RNAs and is located in the dense fibrillar component (DFC) of the nucleolus. The encoded protein contains an N-terminal repetitive domain that is rich in glycine and arginine residues, like fibrillarins in other species. Its central region resembles an RNA-binding domain and contains an RNP consensus sequence. Antisera from approximately 8% of humans with the autoimmune disease scleroderma recognize fibrillarin. Fibrillarin antibody [HL2588] detects Fibrillarin protein at nuclear envelope and nucleus by immunofluorescent analysis.
Sample: HeLa cells were fixed in 4% paraformaldehyde at RT for 15 min.
Green: Fibrillarin stained by Fibrillarin antibody [HL2588] (GTX639039) diluted at 1:500.
Red: alpha Tubulin, a cytoskeleton marker, stained by alpha Tubulin antibody [GT114] (GTX628802) diluted at 1:1000.
Blue: Fluoroshield with DAPI (GTX30920).

Various whole cell extracts (30 μg) were separated by 12% SDS-PAGE, and the membrane was blotted with Fibrillarin + FBLL1 antibody [HL2588] (GTX639039) diluted at 1:1000. The HRP-conjugated anti-rabbit IgG antibody (GTX213110-01) was used to detect the primary antibody.
